What is a Generic Drug?

A generic drug is a copy of the brand-name drug with the same dosage, safety, strength, quality, consumption method, performance, and intended use. Description

Afinitor (everolimus) is a prescription medication used to treat the following:

  • In postmenopausal women: Advanced hormone receptor-positive, HER2-negative breast cancer in combination with exemestane after treatment failure with letrozole or anastrozole
  • In adults: Progressive neuroendocrine tumors of pancreatic origin and progressive, well-differentiated, non-functional neuroendocrine tumors of lung or gastrointestinal origin that are unresectable, locally advanced, or have spread to other parts of the body
  • In adults: Advanced kidney cell cancer after treatment failure with sunitinib or sorafenib
  • In adults: Kidney angiomyolipoma and tuberous sclerosis complex (TSC) that do not require surgery immediately
  • TSC in adults and pediatric patients who are at least 1 years old with subependymal giant cell astrocytoma requiring therapeutic intervention but cannot be curatively resected
  • TSC-associated partial-onset seizures in adults and pediatric patients who are at least 2 years old as an adjunctive treatment

Afinitor comes in 2.5 mg, 5 mg, 7.5 mg, and 10 mg tablets for oral administration.

Directions

Afinitor should be taken as your doctor has prescribed. Your treating physician will determine an appropriate regimen for you.

Afinitor should be kept between 20°C and 25°C (68°F and 77°F); keep out of light and moisture.

Ingredients

Everolimus is the active ingredient in Afinitor.

Anhydrous lactose, butylated hydroxytoluene, crospovidone, hypromellose, lactose monohydrate, and magnesium stearate are the inactive ingredients.

Contraindications

Afinitor should not be administered to patients who are allergic to everolimus or other rapamycin derivatives.

Cautions

  • Before you begin treatment with Afinitor, inform your doctor of the following:
    • All the prescription and nonprescription medications you take
    • Your allergies
    • Your current health conditions
    • Your past medical history
    • Your pregnancy or breastfeeding status
  • Afinitor is associated with the following warnings and precautions:
    • Non-infectious pneumonitis
    • Infections
    • Severe allergic (hypersensitivity) reactions
    • Swelling, or angioedema
    • Inflammation and swelling of the mouth
    • Kidney failure
    • Risk of impaired wound healing
    • Adverse reactions in elderly patients
    • Metabolic disorders
    • Bone marrow suppression
    • Risk of infection or decreased immune response with vaccination; live vaccines and close contact with people who have had live vaccines should be avoided
    • Severe radiation reactions
    • Harm to an unborn baby; effective contraception is recommended during treatment and for 8 weeks after taking the final dose of treatment in females of reproductive potential; effective contraception is recommended during treatment and for 4 weeks after taking the final dose of treatment in male patients with female partners

Side Effects

Afinitor side effects may include but are not limited to inflammation and swelling on the mouth, infections, rash, fatigue, diarrhea, swelling, abdominal pain, nausea, fever, weakness, cough, headache, and decreased appetite.
