Orladeyo (Berotralstat) is a prescription drug used to prevent attacks of hereditary angioedema (HAE) in individuals aged 12 and older. As a plasma kallikrein inhibitor, Orladeyo works by blocking the activity of kallikrein, which reduces the levels of bradykinin, a chemical that causes pain and swelling in HAE patients. This medication is available in capsule form. It is not indicated for the treatment of acute HAE attacks.
To take Orladeyo, follow your doctor's instructions. The standard dosage is typically one 150 mg capsule taken once daily with food to prevent HAE attacks. For people with moderate or severe liver impairment, a reduced dosage is recommended. Always swallow the capsule whole; do not crush, chew, or open it.

Taking more than one capsule of Orladeyo a day may lead to serious complications, such as QT prolongation—a heart rhythm condition that can alter the heart's normal beat. Rare side effects include liver function changes and occasional, brief itchy rashes.
