What is a Generic Drug?

Description

Provera (medroxyprogesterone acetate tablets, USP) is a medication prescribed for treating secondary amenorrhea and abnormal uterine bleeding caused by hormonal imbalance in the absence of organic pathology (i.e., uterine cancer or fibroids).

Provera is available in 2.5 mg, 5 mg, and 10 mg tablets for oral administration.

Ingredients

Medroxyprogesterone acetate is the active ingredient in Provera.

Calcium stearate, corn starch, lactose, mineral oil, sucrose, and talc are the inactive ingredients included. The 2.5 mg tablets also contain FD&C Yellow No.6, and the 5 mg tablets also contain FD&C Blue No.2–Aluminum Lake.

Contraindications

Provera should not be taken by patients with any of the following:

  • Genital bleeding that is abnormal and not diagnosed
  • Current, suspected, or history of breast cancer
  • Estrogen- or progesterone-dependent abnormal growth that is known or suspected
  • Current or history of blood clots in the veins or lungs
  • Current or history of arterial thromboembolic disease such as heart attack or stroke
  • Known angioedema or anaphylactic reaction to Provera
  • Liver disease or dysfunction
  • Pregnancy that is known or suspected

Cautions

  • Before you start taking Provera, tell your doctor about:
    • All the medications you take, including prescription and nonprescription medications
    • Your allergies
    • Your current health conditions
    • Your previous medical history
    • Your pregnancy or breastfeeding status
  • Provera is associated with the following Boxed Warning:
    • Estrogen plus progestin should not be used to prevent cardiovascular disease or dementia.
    • An increased risk of deep vein thrombosis (DVT), pulmonary embolism (PE), heart attack, and stroke in postmenopausal women (aged 50-79 years) during 5.6 years of treatment with daily oral conjugated estrogens (CE) combined with medroxyprogesterone acetate (MPA), relative to placebo, was reported in the Women’s Health Initiative (WHI) estrogen plus progestin substudy.
    • An increased risk of developing probable dementia in postmenopausal women (aged 65+) during 4 years of treatment with daily CE combined with MPA, relative to placebo, was reported in a WHI Memory Study (WHIMS) estrogen plus progestin ancillary study.
    • An increased risk of invasive breast cancer was demonstrated in a WHI estrogen plus progestin substudy.
    • These risks should be assumed to be similar for other CE and MPA doses and other dosage forms and combinations of estrogens and progestins in the absence of comparable data.
    • The lowest effective doses of progestins with estrogens at the shortest duration should be prescribed with considerations of treatment goals and risks for each individual woman.
  • Provera is associated with the following warnings:
    • Cardiovascular disorders: increased risk of blood clots in the veins and lungs, heart attack, and stroke
    • Coronary heart disease
    • Abnormal growths that are malignant (breast cancer, endometrial cancer, ovarian cancer)
    • Probable dementia
    • Vision problems
    • Vaginal bleeding that is abnormal and unexpected
    • High blood pressure
    • High triglyceride levels
    • Liver dysfunction and/or history of cholestatic jaundice
    • Fluid retention
    • Low calcium levels in the blood
    • Altered lab results
    • Exacerbation of asthma, diabetes, epilepsy, migraine, porphyria, systemic lupus erythematosus, and liver hemangiomas

Side Effects

Provera side effects may include but are not limited to menstrual changes, breast tenderness, nausea, vision changes, depression, sleep problems, dizziness, headache, and hypersensitivity reactions.
