What is a Generic Drug?

Description

Sporanox (itraconazole) Oral Solution is prescribed to treat fungal infections of the mouth, throat, and esophagus.

Sporanox Oral Solution 150 mL glass bottles contain 10 mg of itraconazole per mL.

Sporanox Oral Solution should not be used interchangeably with Sporanox capsules.

Directions

Sporanox Oral Solution should be taken as your doctor has prescribed, who will determine an appropriate Sporanox Oral Solution dosage for you. Vigorously swish the solution (10 mL at a time) in the mouth for several seconds, then swallow. Your doctor will tell you how much medication should be used and for how long.

Keep Sporanox Oral Solution stored at or below 25°C (77°F) and out of children’s reach; avoid freezing.

Ingredients

Itraconazole is the active ingredient in Sporanox Oral Solution

Hydroxypropyl betadex, hydrochloric acid, propylene glycol, water, sodium hydroxide, saccharin sodium, sorbitol, cherry, and caramel are the inactive ingredients in Sporanox Oral Solution.

Contraindications

Patients with congestive heart failure (CHF) or a history of CHF should not receive Sporanox Oral Solution unless the infection is serious or life-threatening.

Patients should not take CYP3A4 substrates (for example - not an exhaustive list: methadone, quinidine, ergotamine, irinotecan, lurasidone, oral midazolam, lovastatin, simvastatin, ticagrelor) with Sporanox Oral Solution. Consult your doctor or pharmacist about whether or not you take CYP3A4 medications.

Patients with kidney or liver impairment should not take colchicine, fesoterodine, or solifenacin with Sporanox Oral Solution. Consult your doctor or pharmacist about whether or not you take these medications.

Patients who are poor or intermediate metabolizers of CYP2D6 or are taking strong or moderate CYP2D6 inhibitors should not take eliglustat with Sporanox Oral Solution. Consult your doctor or pharmacist about whether or not you take these medications.

Patients with chronic lymphocytic leukemia (CLL) or small lymphocytic lymphoma (SLL) should not take venetoclax with Sporanox Oral Solution during dose initiation or ramp-up phase of venetoclax.

Patients with an allergy to itraconazole should not take Sporanox Oral Solution. Patients who are hypersensitive to other azoles should use Sporanox Oral Solution with caution.

Cautions

  • Sporanox Oral Solution contains the following FDA Boxed Warning:
    • The continued use of Sporanox Oral Solution should be reassessed if signs or symptoms of CHF occur during administration.
    • Negative inotropic effects were seen in dogs and healthy human volunteers when intravenous (IV) itraconazole was administered to them.
    • Methadone, disopyramide, dofetilide, dronedarone, quinidine, isavuconazole, ergot alkaloids (such as dihydroergotamine, ergometrine (ergonovine), ergotamine, methylergometrine (methylergonovine)), irinotecan, lurasidone, oral midazolam, pimozide, triazolam, felodipine, nisoldipine, ivabradine, ranolazine, eplerenone, cisapride, naloxegol, lomitapide, lovastatin, simvastatin, avanafil, ticagrelor, finerenone, voclosporin, and other CYP3A4 substrates should not be taken with Sporanox Oral Solution.
    • Patients with kidney or liver impairment should not take colchicine, fesoterodine, and solifenacin with Sporanox Oral Solution.
    • Patients who are poor or intermediate metabolizers of CYP2D6 or are taking strong or moderate CYP2D6 metabolizers should not take eliglustat with Sporanox Oral Solution.
    • Patients with CLL or SLL should not take venetoclax with Sporanox Oral Solution during initiation and ramp-up phase of venetoclax.
    • Taking Sporanox Oral Solution with these medications can increase their blood concentrations, which can result in increased or prolonged pharmacologic effects and/or adverse reactions, such as QT prolongation and ventricular tachyarrhythmias like torsades de pointes, which can be fatal.
  • Before you begin treatment with Sporanox Oral Solution, inform your doctor of:
    • All the prescription and nonprescription medications you take
    • Your allergies
    • Your current health conditions
    • Your past medical history
    • Your pregnancy or breastfeeding status
  • Sporanox Oral Solution contains the following warnings and precautions:
    • Liver toxicity, liver failure, and death
    • Heart arrhythmias
    • Heart disease
    • Pseudoaldosteronism
    • Drug interactions
    • Hydroxypropyl-ß-cyclodextrin, an excipient in Sporanox Oral Solution, has produced adenocarcinomas in the large intestine and exocrine pancreatic adenocarcinomas in a rat carcinogenicity study
    • Not recommended for treatment initiation in patients at immediate risk of systemic fungal infections
    • Neuropathy
    • Switching to an alternative therapy should be considered in patients with cystic fibrosis who do not respond to Sporanox Oral Solution
    • Hearing loss that can be transient or permanent

Side Effects

Sporanox Oral Solution side effects may include but are not limited to nausea, diarrhea, vomiting, abdominal pain, constipation, fever, chest pain, pain, fatigue, coughing, shortness of breath, pneumonia, sinusitis, increased sputum, rash, increased sweating, skin disorder, headache, dizziness, pneumocystis carinii infection, and depression.
